Distribution of Adenosine Triphosphatase Activity in Seminiferous Tubules during Spermatogenesis in Mice




Abstract
Spermatogenesis is the process by which a spermatogonial stem cell gives rise to spermatozoan in the selniniferous tubule of the mammals. Many histologist had already interpreted the static histological findings of seminiferous epithelium at various stages of spermatogenesis. It is well known that the various types of evolving germ cells, i.e., the spermatogonia, primary and secondary spermatocytes, and spermatid at various steps of their development are arranged with well defined cellular association.
In this experiment, the author investigated in changes in the distribution of adenosine triphosphatase activity in seminiferous epithelium of mouse at various ages.
Healthy male mice (Balb-C strain) aged from 1 days to 35 days were used as experimental animals and given water and food ad libitum. The experimental animals were sacrificed at 1st, 7th, 14th, 21st, and 35th day of age and testis specimens were obtained.
The specimens sectioned by the cryostat were stained by Wachstein and Meisel method for studying changes in distribution of adenosine triphosphatase activity. Also stained were the specimens with the hematoxylin-eosin.
The results obtained were as follows.
1. Basal layer of selniniferous epithelium showed weak to moderate activity of adenosine triphosphatase in neonatal mouse and revealed weak activity of adenosine triphosphatase in mature mouse.
2. Intermediate and inner layer of seminiferous epithelium showed weak activity of adenosine triphosphatase in neonatal mouse and revealed moderate to strong activity of adenosine triphosphatase in mature mouse.
It is consequently suggested that adenosine triphosphatase activity of intermediate and inner layer of seminiferous epithelium is increased with the progression in differentiation of spermatogenic cells.
